Escuela de Doctorado K1 Huesos - Regeneración K1 Tissue engineering K1 Ingeniería tisular K1 Stem cells K1 Células madre K1 32 Ciencias Médicas AB A new inclusion and cutting method has been developed applied to a bone regeneration study in which the suitability of the mentioned method has been assessed, as well as the regeneration performed on rat mandibular bone after the performance of a critical bone defect (4 mm) in said bone, observing, at all times, the European and Spanish legislation for the use and management of laboratory animals.Four groups of animals were established: A) Positive Control: Subcritical osteotomy (2-3 mm); B) Negative Control: Critical osteotomy (4 mm) without implant or added cells; C) Critical osteotomy with perforated titanium membrane implant; D) Critical osteotomy with the same implant of perforated titanium membrane seeded with adipose-derived mesenchymal stem cells.Animals were sacrificed 30 days after surgery.The new inclusion and cutting method that we have developed and called “the three sections” offers great advantages for obtaining serial sections of hard tissues and large pieces and being very suitable to study them at different levels of magnification, allowing a wide variety of histological stains to be performed on these sections. In this thesis we performed the techniques of toluidine blue, alizarin red, Von Kossa, Fluoropaque, transmission electron microscopy, scanning electron microscopy, and microanalysis by energy dispersive X-rays (EDAX).The experimental study of the animals subjected to osteotomy revealed that group A (subcritical osteotomy) completely regenerated the bone, those in group B (negative control) did not show regeneration, while those in groups C and D (implanted with titanium membrane) were able to regenerate the bone after one month from the osteotomy, with a more complete regeneration in group D due to the effect of the added stem cells. The scanning electron microscope showed a trabecular structure of the newly formed bone and EDAX microanalysis showed hydroxyapatite in its matrix.
